Protective effect of Coix biopeptide on human umbilical vein endothelial cells injured by high glucose

Abstract: To investigate the protective effect and mechanisms of coix biopeptide on high glucose injured human umbilical vein endothelial Cells (HUVEC). The injury model of HUVEC was established by the treating the HUVEC with 30 mM glucose, and then the protection effect of the Coix biopeptide was evaluated. Cell viability was detected by MTT assay. Nitric acid reductase was used to detect NO secretion. The content of reactive oxygen species (ROS) was detected by flow cytometry. Apoptosis and necrosis were detected by Annexin V-FITC/PI double staining. Quantitative real-time PCR was applied to detect the expression of inflammation-related genes. The results showed that NO secretion decreased by 28.72% (P < 0.05), ROS secretion increased by 96.19% (P < 0.05), the expression of ACE gene was significantly decreased, and the expression of ICAM-1, IL-6, MCP-1 and VCAM genes was significantly increased in HUVEC induced by high glucose injury. With the prolongation of high glucose treatment, the proportion of apoptotic and necrosis cells in HUVEC increased significantly, which was 1.61 times of that in NG group. Coix biopeptides increased NO secretion and decreased ROS content to varying degrees, reversed the decrease in ACE gene expression caused by high glucose injury, significantly reduced the expression of inflammatory genes and reduced cell apoptosis. In summary, Coix biopeptide possessed explicit protection effect on by high glucose injured HUVEC.
