Iridoid constituents and anti-aging activity of Eucommiae Folium

Abstract: Eucommiae Folium, a leaf of the genus Eucommia in the family Eucommiaceae, this study focused on the iridoid compounds in medicine food homology plant Eucommiae Folium and explored their potential applications in the field of anti-aging. Nine iridoid compounds, BuA (1), (2) 6-0-Caffeoylajugol, (3) 6-O-(E)-feruloylajugol,6-O-(E)-caffeoylaucubin (4), Specioside (5), Acuminatuside (6), Iso-Scrophulariosid (7), 6-O-acetylscandoside (8), Aucubin (9) were isolated from Eucommiae Folium, Through modern separation techniques, including column chromatography on silica gel column (CC), ODS, Sephadex LH-20 and semi-preparative HPLC. Their structures were elucidated on the basis of spectroscopic and chemical data, especially NMR and MS. Compound 1 was a new iridoid, and compounds 2-7 were isolated from the Eucommiae Folium for the first time. Tests on the anti-aging activity of Caenorhabditis elegans showed that compounds 1, 4, 6, 8, and 9 were found to significantly prolong the lifespan of Hidradenitis elegans nematode, compounds 1, 4, 5, and 9 significantly improved the resistance to oxidative stress, and compounds 1, 8, and 9 significantly improved the resistance to heat stress. These results indicated that the iridoid compounds of Eucommiae Folium have significant anti-aging effects, which provide a scientific basis for future in-depth research and development.
