非衍生化HPLC法分析银耳多糖中单糖组成的初步研究

摘要: 本研究采用非衍生化高效液相色谱法,以分离度与峰形两个因素作为评价指标,建立了鼠李糖、葡萄糖、果糖、木糖、半乳糖五种常见单糖的最佳分离条件。并应用所建立的方法对银耳多糖中的单糖组成进行了分析。初步确定银耳多糖中含有鼠李糖、木糖、果糖和葡萄糖,摩尔比为0.29:1.00:1.01:1.78。为此法在多糖的单糖组成测定中的更好运用提供了依据。

Abstract: This article is discussing a simple and sensitive high performance liquid chromatographic method(HPLC) for the analysis of monosacchoride composition in polysaccharides. Five monosaccharides are separated by non-derivatization HPLC with RI monitor. By means of HPLC analysis rhamnose, glucose, fructose, xylose, galactose five kinds of monosaccharides, the best separation condition of the monosaccharide is obtained. The monosaccharide composition of Tremella polysaccharide is determined by a data analysis method which is developed to ensure accuracy and reproducibility.
