北五味子木脂素对酒精性肝损伤的保护作用

Protective Effect of Schisandra Lignans on Liver Injury Induced by Ethanol

摘要: 目的 研究北五味子木脂素(Schisandra Lignans,SCL)对酒精诱导小鼠急性肝损伤的保护作用并初步探讨其作用机制。方法 将50只小鼠随机分为正常对照组、模型组、SCL低剂量组(25 mg/kg)、SCL高剂量组(50 mg/kg)、阳性对照组(联苯双酯150 mg/kg),灌胃给药预处理15天。末次给药1h,造模组小鼠给予50%乙醇12mL/kg一次性灌胃,正常对照组给予同体积蒸馏水。12h后处死小鼠,检测血清中谷丙转氨酶(alanine aminotransferase, ALT)和谷草转氨酶(aspartate aminotransferase, AST)活性,甘油三脂(triglyceride, TG)水平,肝组织中丙二醛(malondialdehyde, MDA),还原型谷胱甘肽(glutathione, GSH)含量及一氧化氮合酶(nitric oxide synthase, NOS)活性;HE染色观察肝脏病理学改变。结果 SCL可明显降低酒精致肝损伤小鼠血清ALT和AST活性以及TG水平(P<0.05),同时使小鼠肝组织MDA含量和NOS活性显著下降,GSH含量提高(P<0.05);HE染色结果显示SCL可显著改善酒精引起的小鼠肝细胞水肿、坏死,中央静脉充血等病理损伤。结论 SCL对酒精性肝损伤具有明显的保护作用,其机制可能与抗氧化有关。

Abstract: Objective To explore the protective effect of Schisandra lignans (SCL) on acute liver injury induced by ethanol in mice and to reveal its mechanism. Methods 50 mice were randomly divided into normal control group, model group, low-dose SCL group (25 mg/kg), high-dose SCL group (50 mg/kg) and positive control group (bifendate, 150 mg/kg). The corresponding agents were intragastrically administrated to all the mice respectively for 15 days. 50% ethanol (12.0 mL/kg) was given to the mice in model groups 1 hour after the last administration. The mice of normal control group were administrated match volume distilled water. All the mice were sacrificed 12 hours later, and their blood samples were collected. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities, triglyceride (TG) level in serum, malondialdehyde (MDA) and reduced glutathione (GSH) contents, and nitric oxide synthase (NOS) activity in liver tissues were measured. HE staining was performed for observing pathological changes of the liver tissues. Results SCL could significantly decrease the ALT, AST activities and TG levels in serum(P<0.05). Meanwhile the MDA contents and NOS activities in liver tissues were reduced significantly, and the GSH contents in liver tissues were increased by SCL(P<0.05). Moreover, HE staining showed that SCL could significantly improve the liver cell edema, necrosis, and central venous congenstion. Conclusion Schisandra lignans has a significant protective effect on alcohol-induced acute liver injury, which might be related to the anti-oxidation.
