杜氏藻中beta-胡萝卜素的水溶性有机溶媒萃取

Beta-carotene extraction from Dunaliella salina by water-soluble organic media

摘要: 摘要:本项研究的目的是使用水溶性有机溶媒建立适用于杜氏藻中?-胡萝卜素萃取的方法。在实验中,以乙酸乙酯为参比,用丙酮、甲醇、乙醇和四氢呋喃4种水溶性有机溶媒,对实验室条件下养殖的杜氏藻体在室温条件下分别进行萃取。用C18-HPLC和C30-HPLC分别测定萃取物中?-胡萝卜素含量及其几何异构体的组成。通过比较不同萃取溶媒的?-胡萝卜素萃取得率和所消耗的溶媒体积,筛选出适合食品工业生产中使用的水溶性萃取溶媒。在进一步考虑各溶媒的食品安全性之后,确定乙醇为最适溶媒。使用乙醇作为萃取溶媒,能够从每毫克(干重)藻体中萃取得到?-胡萝卜素0.32微克。其中,总顺式异构体的比例占20.21%(W/W)。随后,使用4种碳水化合物和1中蛋白质水解酶对藻体的细胞壁和膜进行破坏。观察原料酶解对?-胡萝卜素萃取率和溶媒消耗的影响。实验结果表明:原料酶解可导致其中?-胡萝卜素的损失,并对溶剂消耗无明显改进,不建议采用。

Abstract: Abstract: This study aims to develop a method of ?-carotene extraction from Dunaliella salina by water-soluble organic solvents. ?In experiments, in refer to ethyl acetate, four water-soluble organic solvents, acetone, methanol, ethanol and tetrahydrofuran, were selected as as media to extract ?-carotene from Dunaliella salina cultured under lab-conditions. ?-carotene amount and its geometrical composition were determined by C18-HPLC and C30-HPLC, respectively, of each extract. By the ?-carotene extracting recovery and solvent consumption comparisons of each solvent, proper extracting media were selected for industrial practice. In further refer to food safety, ethanol was selected as the most proper medium. By ethanol as extracting medium, 0.32 ?g of ?-carotene was extracted from 1 mg of dried Dunaliella salina with 20.21% (W/W) of total Z-isomers. Four carbohydrate and a protein hydrolysis enzymes were then employed to destroy the cell wall and membrane of algae. The influences of raw material enzymatic hydrolysis on ?-carotene extracting recovery and solvent consumption were observed. Data from this investigation suggested that raw material enzymatic hydrolysis resulted in the loss of ?-carotene amount and exhibited no improvement in solvent consumption. It is therefore not considered to be applied.
