桑叶生物碱对氧化应激小鼠糖脂代谢及肝损伤的改善作用

Improvement Effect of mulberry alkaloids on Abnormal Glucose and Lipid Metabolism and Liver Injury Induced by Oxidative Stress

摘要: 目的:探究桑叶生物碱对氧化应激所致实验小鼠糖脂代谢异常及肝损伤是否具有改善作用,旨在为桑叶生物碱的深入利用提供理论依据。方法:采用腹腔注射1000 mg/kg?BW D-半乳糖建立小鼠氧化损伤模型,分别给造模成功的实验小鼠灌胃低(50 mg/kg?BW)、中(100 mg/kg?BW)、高剂量(200 mg/kg?BW)的桑叶生物碱及抗氧化剂还原型谷胱甘肽(GSH,Glutathione,200 mg/kg?BW),于0、4、8、12周末取样,测定各组小鼠空腹血糖、血脂水平及肝损伤指标的变化。结果:在本研究实验周期及剂量范围内,与模型对照组相比,阳性药物组和桑叶生物碱各剂量组小鼠的空腹血糖水平、血脂胆固醇(total cholesterol,TC)、甘油三酯(triglycerides,TG)水平,肝损伤指标谷丙转氨酶(alaninetransaminase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)水平均显著下降(P<0.05)。同时,桑叶生物碱高剂量组与阳性药物组之间结果无显著差异(P>0.05),且均恢复到正常水平(P>0.05),它们对小鼠糖脂代谢异常及肝损伤的改善作用呈现较明显的剂量依赖性,但无时间效应。肝脏组织切片观察结果也显示阳性药物组和桑叶生物碱各剂量组的肝脏病变程度明显减轻。结论:桑叶生物碱可以改善氧化应激所致小鼠的糖脂代谢异常和肝损伤,其机制可能与抑制氧化应激的发生发展有关。

Abstract: Objective: In order to provide a theoretical basis for the scientific utilization of mulberry leaf, it is explored whether mulberry leaf alkaloids can improve the abnormal glucose and lipid metabolism and liver injury induced by oxidative stress. Method: Two hundred and twenty male Kunming mice were randomly divided into six groups, model group, positive control group (200 mg/kg of glutathione) and mulberry alkaloids low-dose, medium -dose and high-dose (50,100, 200 mg/kg?BW). To establish a oxidative damage model by using intraperitoneal 1000 mg/kg?BW D-galactose for 20d. The changes of fasting blood glucose, lipid level and liver injury were measured at weekends of 0, 4, 8 and 12. Results: In the experimental period and dose range of this study, com- pared with model group, the fasting blood glucose, the total cholesterol (TC), triglycerides (TG), the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) were significantly decrease in the positive control group and three dose of mulberry alkaloids groups(P<0.05). Meanwhile, there was no significant difference between the high dose group of mulberry alkaloids and the positive drug group, and both recovered to the normal level (P>0.05). The results of histopathological showed that the degree of liver lesions of positive control group and three mulberry alkaloids doses were significantly reduced. Moreover, the effect of mulberry leaf alkaloids and positive drugs on the abnormal glucose and lipid metabolism and liver injury in mice was dose-dependent but had no time effect. Conclusion: Mulberry alkaloids can improve glucose and lipid metabolism abnormalities and liver damage caused by oxidative stress, and its mechanism may be related to inhibiting the occurrence and developmen -t of oxidative stress.
