重金属镉离子单克隆抗体的制备

摘要: 重金属离子镉与螯合剂IEDTA螯合后再与载体蛋白KLH偶联获得完全抗原(Cd-IEDTA-KLH),以该完全抗原免疫Balb/C小鼠,制备杂交瘤获得了稳定分泌抗Cd-ITCBA抗体的细胞株1F6。以该细胞株免疫小鼠腹腔,获得抗Cd-ITCBA的腹水型单克隆抗体,效价为1:51200。

Abstract: Heavy metal ion cadmium was chelated with Isothiocyanobenzyl-EDTA (IEDTA) and linked with vector protein KLH to gain the completed antigen (Cd-IEDTA-KLH). Several Balb/C mouse were immunized with Cd-IEDTA-KLH to make hybridoma cells. The strain 1F6 which can produce monoclonal antibody against Cd-IEDTA steadily was obtained. The ascites was obtained by immunized mouse with 1F6 cells. The titer of the monoclonal antibody in the ascites was about 1:51200.
