产辅酶Q10海洋酵母的筛选及培养条件的优化

摘要: 从50株海洋酵母中选取11株辅酶Q10产量相对高的菌株,经过复筛得到辅酶Q10高产菌株FH-08,经过对氮源、碳源、初始pH值、培养温度等条件的研究分析,得到最佳的发酵条件。通过优化实验,确定培养基的碳源为葡萄糖3%,氮源为酵母粉2%;pH6.5、温度30℃、接种量4%,300ml三角瓶中的装液量为50ml,摇床转速为200r/min。按此条件进行培养,发酵液中菌体生长量为2.34g/200ml,辅酶Q10的产量为28.6mg/L。

Abstract: 11 coenzyme Q10-producing strains were isolated from 50 marine yeasts preliminarily. A high-producing strain,FH-8 was obtained through rescreening from those 11 yeasts. The fermentation conditions of CoQ10 by FH-08 were optimized. The results showed that the optimal fermentation conditions are as following:carbon source of glucose 3%,nitrogen source of yeast extract 2%,initial pH 6.5,inoculum size 4%,filled volume of medium 50 ml/300 ml,temperature 30℃ and rotation speed 200 r/min. Under these fermentation conditions,the biomass reaches 2.34 g/200 ml,and the CoQ10 concentration in broth reaches 28.6 mg/L.
