苹果多酚HPLC检测条件的优化研究

摘要: 本实验高效液相色谱以色谱柱为C18柱(150mm×4.6mm,5μm),流动相为甲醇和醋酸,柱温为40℃和20℃,流速为1.00ml/min,线性梯度洗脱,紫外检测器波长为280nm和320nm。在此色谱条件下,各组分均得到良好分离。用乙酸乙酯提取苹果中的酚类物质,在pH值为2.0时分离提取酸性酚,在pH值为7.0时分离提取中性酚,通过对中性酚和酸性酚色谱分离条件的摸索和优化,最终选用RP-C18色谱柱,中性酚的分离以4%醋酸缓冲液和甲醇为流动相,酸性酚的分离以甲醇-4%醋酸为流动相,建立起酚类物质高效液相色谱测定方法。

Abstract: High performance liquid chromatography(HPLC) was used to detect apple polyphenols,and its basic operation conditions were determined as:C18 column(150×4.6 mm,5 μm),methanol-acetic acid as mobile phase,flow rate 1.00 ml/min,column temperature 40 ℃ and 20 ℃ and wavelength of detector 280 nm and 320 nm. All compositions of apple polyphenols were separated completely under these chromatography conditions. Phenolic compounds were extracted from apples with ethyl acetate. Neutral phenolics were extracted at pH 7.0 and acid phenolics were extracted at pH 2.0. RP-C18 column is suitable to separate apple polyphenols and the mobile phase for the separation of neutral and acid phenolics is identical,the mixture of 4% acetic acid and purified water.
