葡萄籽和皮萃取物对小鼠乳腺癌细胞迁移的影响

Effects of Grape Seed and Grape Skin Extracts on the Migration of Mouse Mammary Carcinoma Cells

摘要: 目的:研究葡萄籽和皮萃取物对乳腺癌细胞迁移的抑制作用,以及对细胞活力的影响。方法:以小鼠乳腺癌细胞4T1为研究对象,四甲基偶氮唑盐(MTT)实验检测细胞增殖活性;细胞迁移实验检测细胞迁移能力;结果:质量浓度为5μg/mL的葡萄籽和皮萃取物对乳腺癌细胞的迁移具有明显的抑制作用,该抑制作用与药物质量浓度呈现剂量依赖关系;萃取物对癌细胞的活力有明显的抑制作用,葡萄籽和皮萃取物的半数有效质量浓度(IC50)分别为65.427μg/mL和72.408μg/mL。

Abstract: Objective: To explore the effects of grape seed and grape skin extracts on the migration, viability and proliferation of breast cancer cells. Methods: The viability of mouse mammary carcinoma 4T1 cells was determined by MTT assay and the migration ability by transwell chemotaxis assay. Results: Both grape seed and grape skin extracts significantly reduced the migration of the 4T1 cells in a dose-dependent manner, and exhibited an obvious inhibitory effect on cell migration at a concentration as low as 5μg/mL. The extracts also obviously inhibited the viability and proliferation of the 4T1 cells with IC50 (half maximal inhibitory concentration) of 65.427μg/mL and 72.408μg/mL, respectively.
