2-羧基苯偶氮若丹宁固相萃取光度法测定生物样品中的铅

摘要: 研究了2-羧基苯偶氮若丹宁(CPAR)与铅的显色反应,在pH4.0的盐酸-六次甲基四胺缓冲介质与乳化剂OP存在下,CPAR与铅反应生成2:1稳定络合物,可被Waters Sep-PakC18小柱固相萃取,用氮-氮二甲基甲酰氨(DMF)洗脱后于520nm处光度法测定,体系摩尔吸光系数ε为6.75×104L/mol·cm,铅含量在0.05~4.0μg/ml内符合比尔定律,本方法可用于生物样品中铅含量的测定,结果令人满意。

Abstract: The color reaction of lead with 2-carboxyphenylazo-rodanine (CPAR) was studied. In the presence of pH4.0 buffer solution and emulsifier-OP medium, CPAR could react with lead(II) to form a stable 2:1 complex. This complex extracted by Waters Sep-Pak-C18 cartridge and eluted by DMF, its spectrophotometry determination results showed that the molar absorptivity is 6.75×104 L/mol·cm at 520 nm. Beer’s law was obeyed in range of 0.05~4 μg/ml. This method can be applied for the determination of lead in biological sample with satisfactory results.
