壳寡糖对小鼠急性酒精性肝损伤的保护作用

Protective Effect of Chitooligosaccharides on Acute Alcohol-Induced Liver Injury in Mice

摘要: 本实验研究了壳寡糖对急性酒精性肝损伤的保护作用。建立小鼠急性肝损伤模型,连续灌胃受试物(壳寡糖)30 d后给予12 mL/kg mb的体积分数50%乙醇溶液后处死,对小鼠进行了肝脏病理组织学检查,并测定了血清中谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)活力,总胆固醇(total cholesterol,TC)浓度,以及肝脏系数、肝脏中谷胱甘肽(glutathione,GSH)含量、超氧化物歧化酶(superoxide dismutase,SOD)活力和丙二醛(malondialdehyde,MDA)含量。结果显示,与酒精模型组相比,壳寡糖组小鼠肝组织损伤程度有所减轻,阳性对照组和壳寡糖组血清ALT、AST和TC水平显著降低,肝脏GSH含量和SOD活力均显著增加,而肝脏系数和MDA含量显著降低(P<0.05),表明壳寡糖对小鼠酒精性肝损伤有明显的保护作用。

Abstract: In this paper, the protective effect of chitooligosaccharides (COS) on acute alcohol-induced liver injury was studied. A mouse model of acute liver injury was established by administering 50% ethanol at a dose of 12 mL/kg mb to mice after 30 continuous days of oral gavage with COS. The mice were sacrificed for histopathological examination of liver tissue. Besides, the activity of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and total cholesterol (TC) concentration in serum, liver coefficient, and glutathione (GSH) content, superoxide dismutase (SOD) activity and malondialdehyde (MDA) content in liver were detected. The results showed that the degree of liver injury in the COS group was mitigated compared with the alcohol model group. The positive control and COS groups exhibited a significant reduction in serum ALT, AST and TC levels and a significant increase in hepatic GSH content and SOD activity as well as a significant decrease in liver index and MDA level (P < 0.05). Therefore, chitooligosaccharides has a significant protective effect on alcohol-induced liver injury in mice.
