RPA、RAA及ERA技术在食源性致病菌快速检测中的研究进展

Research progress of RPA, RAA and ERA techniques in rapid detection of foodborne pathogens

摘要: 随着人民生活水平的提高,食品安全问题也越来越受社会关注,其中生物(微生物)因子是影响食品安全的最主要因素。而由于等温扩增技术不需依赖复杂的仪器设备,能够快速、准确地进行生物成分检测而被广泛应用于食品安全研究,尤其是新发展起来的重组酶聚合酶扩增技术(Recombinase Polymerase Amplification,RPA)、重组酶介导链替换核酸扩增技术(Recombinase Aided Amplification,RAA)和酶促重组等温扩增技术(Enzymatic Recombinase Amplification,ERA)。这三种等温扩增技术相对于其他等温扩增技术具有反应条件更温和,引物设计更简单等优点,且检测原理相似,因此本文对这三种技术进行了对比研究,从原理、概念以及近年来在食源性致病菌快速检测方面的研究应用情况进行综述,概括了其在食品检测中的优势和面临的实际问题,并对未来的发展前景提出了展望。

Abstract: With the improvement of people’s living standards, food safety has attracted more and more social attention. Among of them, the biological (microorganism) factor is the most important factor that affects the food safety. Isothermal amplification technology is widely used for food safety because it can detect biological ingredients rapidly and accurately without relying on complicated instruments, especially the new techniques of recombinase polymerase amplification (RPA), recombinase aided amplification (RAA) and enzymatic recombinase amplification (ERA). Compared with other isothermal amplification techniques, they have the advantages of mild reaction conditions and simple primer design, as well as with similar principles. Therefore, in this paper, these three technologies were introduced, compared, and reviewed the concepts, principles, and the application in the rapid detection of food pathogens in recent years. The advantages and practical problems of them in food inspection were summarized, and the prospects of future development were put forward.
