草菇子实体多肽对小鼠急性酒精肝的预防作用及肠道菌群的影响

Preventive Effects of Volvariella volvacea Fruit Polypeptides in a Mouse Model of Acute Alcoholic Liver Injury and Its Influence on Intestinal Bacteria

摘要: 为了探究草菇子实体多肽(Volvariella volvacea Fruit Polypeptides,VVFP)对急性酒精肝模型小鼠预防作用及其对肠道菌群的影响,以先前获得的VVFP(分子量1-3 KD)为材料,灌胃小鼠,将小鼠随机分为空白组、模型组、阳性对照组、多肽低剂量组、多肽中剂量组、多肽高剂量组,比较各组小鼠的血清和肝脏指标以及组织病理切片,利用16s rDNA基因高通量测序,分析各样品中的微生物菌群的生物多样性以及在门和属水平上的相对丰度。结果表明,VVFP能够极显著降低血清中甘油三酯、总胆固醇、谷丙转氨酶、谷草转氨酶、和肝脏中丙二醛的含量,并且降低肿瘤坏死因子-α、白介素-6炎症因子水平,同时明显提高了肝脏中乙醇脱氢酶、总超氧化物歧化酶和谷胱甘肽过氧化物酶的活性。16s rDNA结果表明,VVFP可以使小鼠肠道菌群中α多样性的chao1指数和observed_species指数显著降低,shannon指数升高,并可通过调节拟杆菌门、厚壁菌门、链霉菌属乳酸杆菌属和弧菌属的丰度减轻小鼠肝脏的损伤程度。因此,VVFP可以减轻酒精引起的肝损伤,并为多肽在功能食品领域的应用提供了药效基础。

Abstract: To investigate preventive effects of the Volvariella volvacea fruit polypeptides (VVFP) in a mouse model of acute alcoholic liver injury and its influence on intestinal bacteria. The previously obtained VVFP (molecular weight 1-3 KD) was used as the material to gavage mice. All mice were randomly divided into blank control group, model group, positive control group, polypeptide low-dose group, polypeptide moderate-dose group and polypeptide high-dose group. The serum indexes, liver indexes, and histopathological sections of mice in each group were compared, and 16s rDNA gene high-throughput sequencing was used to analyze the biodiversity of microbial flora and the relative abundance at the phyla and genus levels in each sample. Results showed that the VVFP could highly significantly reduce the contents of triglycerides, total cholesterol, alanine aminotransferase and aspartate aminotransferase in serum and malondialdehyde in liver. The VVFP could also reduce the levels of tumor necrosis factor- α and interleukin-6 inflammatory factors, and significantly increase the activities of alcohol dehydrogenase, total superoxide dismutase and glutathione peroxidase in the liver. 16 S rRNA sequencing showed that VVFP could significantly reduce the chao1 index and observed species index of α diversity in the intestinal flora of mice, and increase the shannon index. VVFP could also reduce the liver damage of mice by regulating the abundance of Bacteroides, Firmicutes, Streptomyces, Lactobacillus and Vibrio. In conclusion, VVFP can reduce the liver injury caused by alcohol, and provide a pharmacodynamic basis for the application of polypeptides in the field of functional food.
