余甘子与茶多酚、左旋肉碱配伍对高脂饮食诱导大鼠肥胖的预防作用

Preventive effect of Phmllanthi Fructus in combination with tea polyphenols and L-carnitine on high-fat diet-induced obesity in rats

摘要: 目的:研究余甘子、余甘子与茶多酚、左旋肉碱配伍对高脂饮食诱导大鼠肥胖的预防作用。方法:实验组、模型对照组和阳性对照组大鼠给予高脂饲料进行喂养,空白对照组大鼠给予普通饲料进行喂养;同时实验组分别灌胃余甘子、余甘子茶多酚混合物、余甘子左旋肉碱混合物。灌胃结束后称量大鼠体重、体内肾周脂肪、睾周脂肪,检测大鼠血清及肝脏组织中血脂四项水平。测定血清中瘦素、抵抗素和脂联素水平;肝脏内肝酶活性、总蛋白水平及脂肪组织内mRNA的表达情况,观察余甘子、余甘子与茶多酚、左旋肉碱配伍对预防肥胖模型大鼠的减肥情况及肝损伤情况。结果:模型对照组大鼠体重、体内脂肪含量相较于空白对照组有明显增加,且差异具有显著性(p < 0.05)。在余甘子的调节作用下,余甘子干预组与余甘子左旋肉碱配伍干预组大鼠体重显著下降,肾周脂肪和睾周脂肪含量也显著降低,血清和肝脏中总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)水平显著降低,高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)水平显著升高。同时血清中瘦素、抵抗素水平显著降低,脂联素水平升高;肝脏中碱性磷酸酶(Alkaline Phosphatase ,AKP)、天冬氨酸转氨酶(Aspartate aminotransferase ,AST)、丙氨酸转氨酶(Alanine aminotransferase ,ALT)、丙二醛(Malondialdehyde ,MDA)酶活性下降,超氧化物歧化酶(Superoxide Dismutase,SOD)活性增加;脂肪组织内AMPKα-2的表达显著上调。余甘子茶多酚配伍干预组预防肥胖效果并不显著。结论:余甘子能够预防高脂饮食诱导的大鼠肥胖,且单独使用效果优于余甘子与茶多酚或左旋肉碱配伍使用。

Abstract: Abstract: Objective: To study the preventive effects of Phllanthus emblica, Phllanthus emblica with tea polyphenols and L-carnitine on high-fat diet-induced obesity in rats. Methods: The experimental group, the model control group and the positive control group were fed with high-fat diet and the negative control group was fed with normal diet. Meanwhile, the experimental group was gavaged with Phllanthus emblica, Phllanthus emblica and tea polyphenol mixture and Phllanthus emblica and L-carnitine mixture respectively. After gavage, the rats were weighed, and their body weight, perirenal fat and peri-testicular fat were measured, and the levels of lipids in serum and liver tissues were detected. The levels of leptin, resistin and lipocalin in serum; liver enzyme activity, total protein level and mRNA expression in adipose tissue were measured to observe the weight loss and liver injury of rats in the obesity prevention model by combining Phllanthus emblica, Phllanthus emblica with tea polyphenols or L-carnitine. Results: The body weight and body fat content of the rats in the model control group increased significantly compared with the negative control group, and the difference was significant (p < 0.05), indicating that the model of obesity in rats induced by high-fat diet was successfully constructed. Under the modulating effect of Phllanthus emblica, the rats in the Phllanthus emblica intervention group and the Phllanthus emblica L-carnitine combination intervention group showed a significant decrease in body weight, perirenal fat and peritesticular fat content. The levels of total cholesterol, triglyceride and low density lipoprotein cholesterol in the serum and liver decreased significantly, and the level of high density lipoprotein cholesterol increased significantly. At the same time, serum leptin and resistin levels were significantly reduced, and lipocalin levels were increased; the activities of Alkaline Phosphatase, aspartate Aminotransferase, alanine aminotransferase and Malondialdehyde in the liver were decreased, and the activity of Superoxide dismutase was increased; the expression of AMPKα-2 was significantly up-regulated in adipose tissue. The obesity prevention effect was not significant in the Phllanthus emblica, tea polyphenol combination intervention group. CONCLUSION: Phllanthus emblica prevents high-fat diet-induced obesity in rats, and the effect of Phllanthus emblica alone is better than that of Phllanthus emblica in combination with tea polyphenols or L-carnitine.
