Application of Gene Chips in Analysis of Differential Expressions of Genes Involving in Aflatoxin Biosynthesis

Abstract:

The aflatoxin (AF) production was induced in Aspergillus parasiticus AS3.4407 by shifting the cultures from nonconducive to conducive medium and aflatoxin B1 (AFB1)-producing and non-AFB1-producing mycelia were obtained and lyophilized. Subsequently, differential expressions of genes involving in the AF biosynthesis were screened using gene chips and RT-PCR. The results from gene chips were consistent with those from RT-PCR amplification, suggesting differential expressions of six genes including aflA, aflE, aflF, aflR, aflT and aflX. This study proves that the differential expression of aflR is closely related to the aflatoxin production.
