Simultaneous Determination of Phenolic Acids in Potato Tuber by RP-HPLC

Abstract: Objective: A reversed-phase high performance liquid chromatographic (RP-HPLC) method was developed to determine the contents of chlorogenic acid, caffeic acid, p-coumaric acid and ferulic acid in potato tuber from Jiangxi, Beijing and Xinjiang. Methods: The contents of phenolic compounds were measured by RP-HPLC using YMC-C18 column (250 mm × 4.6 mm, 5μm) as the sationary phase and CH3OH: H2O: CH3COOH (30:70:1, V/V) as the mobile phase at a flow rate of 1.0 mL/min. Results: A good linear relationship of this method was achieved in the ranges of 7.80-1000μg/mL for chlorogenic acid, 0.78-100μg/mL for caffeic acid, 0.312-40μg/mL for p-coumaric acid and 0.078-10μg/mL for ferulic acid with a correlation coefficient of 0.9968-0.9999. The average recovery rates of this method were 92.38%-101.73%. Conclusion: The developed RP-HPLC method is characteristic of fast determination, accurate results, high sensitivity and good reproducibility. In addition, different growth environments of potato tuber have an important impact on the contents of phenolic acids.
