Determination of Flavonoids in Extract of Ginkgo biloba L.by High Performance Liquid Chromatography

Abstract: High-performance liquid chromatography (HPLC) was used to evaluate the influence of different variables affecting the hydrolysis of flavonoids in extract of Ginkgo biloba L.(EGb).The hydrolysis recovery was estimated and four kinds of quantiative methods were also been compared.The effects of concentration of HCl in methanol,time and methanol/H2O volume ratio can be interpreed by observing the variation of quantitative results.The acid-hydrolysis conditions optimized by using the orthogonal design method and the anlysis of ranges and variances were obtained as follows:sample soluted in 1.0mol/L HCl-methanol,hydrolyzed at 80℃ for 1.0h,then analyzed by HPLC.Results show that the concentration of HCl and methanol affected the hydrolysis efficiency more markedly than time,difference standard solutions and convert factor cause less imparity to quantiative results.
