Study on High β-1,3-glucanase Strain Screening from Soil and Enzyme Producing Conditions

Abstract: A strain M014, identified as Trichoderma sp., with high β-1,3-glucanase was isolated from the soil. The effects ofthe different nutrients (carbon source and nitrogen source) and cultural conditions (temperature and time) on β-1,3-glucanaseproduction by M014 were studied. The maximal activity of β-1,3-glucanase was reached at 30℃ at 150r/min with 6d incubationin a liquid medium(pH6.0), containing 3% Fuling powder, 0.5% yeast cream and mineral salts. Enzymatic properties of the crudeenzyme filtration from M014 were investigated. The optimal pH and temperature for the enzyme reaction were pH4.5 and60℃, respectively. At the temperature of 40℃ and 50℃ to be preserved for 12h, the activity of β-1,3 glucanase was still stableHowever, over the temperature of 60℃, it descended rapidly.
