Determination of Chloramphenicol Residues in Aquiculture Products by HPLC

Abstract: The residues of chloramphenicol in several kinds of aquiculture products were assayed with the optimized method of high performance liquid chromatography (HPLC). The chloramphenicol in seafood samples was extracted twice by ethyl acetate, and the combined organic phrases were concentrated up to dryness in 60℃ water bath under a gentle stream of nitrogen. Then, its residue was resolved in petroleum-aether and the chloramphenicol re-extracted with mobile phase was filtrated with 0.45μm membrane. The chloramphenicol sample was separated by the RP-HPLC column (discovery, 3.6mm×25cm, 5μm) with a mobile phase of methanol and water (36:65, V/V) at the rate of 1 ml/min and detected at 278 nm. The average recovery was 93.2%, and the detection limit estimated with 3-fold standard deviation was 0.014μg/g. The results showed that chloramphenicol was detected in most of the samples and some of them exceeded the national limitation of China.
