Isolation,Purification and Characterization of the Glycoprotein from Chlorella Pyrenoidosa

Abstract:  The crude glycoprotein was extracted from Chlorella Pyrenoidosa with hot water, and precipitated by ethanol after deproteinization. A refined glycoproteins were prepared after further purification on Sephadex-75 column chromatography, and their homogeneity was examined with Sephadex-200 column chromatography. The ultraviolet spectra showed their characteris- tic absorption peak at 280.00nm and 195.00nm. The molecular weight was estimated to be about 63.7kDa on SDS-PAGE. It was proved that the main monosacchrides were glucose, galactose and xylose by gas chromatography, and O-glycosidic linkage was found by β-elimination reaction.
