Effects of Tomato Juice on Proliferation and Apoptosis of Human Prostate Carcinoma PC-3 Cells

Abstract:
【Abstract】 Objectives: To study the inhibitory effects of tomato juice on the growth of human prostate carcinoma PC-3 cells and the probable mechanism. Methods: PC-3 cells were treated with the tomato juice in different concentrations(40, 80, 120μl/ml) and different contents lycopene for 48h; the proliferation of PC -3 cells were assayed by growth curve and MTT(Thiazolyl blue) assay. TUNEL(Terminal decxynucleotide mediated nick end labeling) were used to observe the morphology of apoptotic cells. The flow cytometry was also used to analyze the PC-3 apoptosis. Results: High concentrations lycopene and tomato juice can markedly inhibit the proliferation of PC-3 cells, while TUNEL positive cells were detectable. Apoptotic peaks by flow cytometry in experimental groups and lycopene group are also detected. The effects measured in experiments above were enhanced with the increasing of tomato juice. The effects of low dose group are weaker than lycopene group but there is no difference between the middle dose group and the lycopene group.The effect of high dose group is stronger than the lycopene group. Conclusions: This study revealed that the anti- prostate cancer effect of tomato juice is not only related to lycopene but also to, other components which have the synergistical interection with lycopene. And lycopene and tomato juice can induce apoptosis of PC-3 cells, a result related to inhibiting the proliferation of PC-3 cells.
