Development of ELISA System to Determine Ochratoxin A(OTA)

Abstract: In the paper, the methods of the indirect competitive ELISA for determining OTA were developed by using self- prepared rabbit serum antibody (IgG) and immunoglublin yolk (IgY). The optimum ELISA parameters with IgY were that the concentration of antigen and blocking agent was 7.5μg/ml, 2.5% and the dilution multiples of IgY and anti-chicken rabbit antibody-HRP were 1000 and 14000, respectively. The optimum parameters with rabbit IgG were that the concentration of antigen and blockaded agent was 2.5μg/ml, 2.5% and the dilution multiples of rabbit IgG and anti-rabbit goat antibody-HRP were 8000 and 10000, respectively. The detection limits of the ELISA with IgY and rabbit IgG were 10ng/ml and 1ng/ml, respectively.
