Determination of Malachite Green in Aquatic Products by HPLC- FLD

Abstract: In this paper, a method for determining the total residue of malachite green in aquatic products was developed by high- performance liquid chromatography (HPLC) with fluorescence detection. Malachite green in sample was reduced to leucomalachite green. Being extracted by acetonitrile-buffer mixture, cleaned-up and condensed, leucomalachite green was determined on a Symmetry shieldTMRP18 column with acetonitrile-water as mobile phase, and monitored with fluorescence detector (λem=265 nm andλem=360 nm). The linear range for leucomalachite green is 1~160 ng/ml. The detection limit of the method according to the total weight of malachite green is 1.0 μg/kg. The recovery is in the range of 82.6%~92.6%.
