Application of SRP-HPLC with Gradient Elution for Rapid Determination of Minocycline Residues in Milk

Abstract:  To quickly establish reverse phase high-performance liquid chromatography (RP-HPLC) method to determine residues of Minocycline in the milk, the gradient elution method was developed. Minocycline was separated on a sunfire C18 column (250mm×4.6mm, 5μm i.d.) with methanol-acetonitrile-0.01mol/L oxalic acid at the flow rate of 1.0 ml/min and detecting at the 355 nm within 8 min. The results showed that the method under the gradient elution is more sensitive and quicker with lower detection limit than that of the isocratic elution. The detection limit is 20μg/kg for Minocycline. The standard curve is obtained in the concentration range of 0.02~1.0μg/ml with the average recovery 99%. The lowest detection limit is 0.02μg/ml. The method established is simple, special, sensitive and reliable. It can be used to detect the residue of Minocycline and the pharmacokinctic parameters of Minocycline in milk.
