Cloning and Bioinformatic Analysis of β-fructofuranosidase Gene from Aspergillus oryzae

Abstract: β-fructofuranosidase (FRU) hydrolyzes sucrose and fructooligosaccharide (FOS). Therefore, the FRU enzymatic activity should be inhibited or eliminated for the production of FOS enzymatically. In this study, a β-fructofuranosidase gene was cloned with RT-PCR from Aspergillus oryzae GX0015 which is a key strain for FOS production industrially. This gene was sequenced and submitted to GenBank (accession number: EU130944). The sequence was analyzed bioinformatically. The results showed that FRU composing of 456 amino acids is a hydrophilic protein without transmembrane helices. The glycoside hydrolysis family 32 domain is present in the 52th to 456th amino acid of FRU and (N/G)DP(C/N)G and RDP conserved sequences are located in the active center of FRU. The phylogenetic tree shows that FRU of Aspergillus oryzae is between yeast invertase and bacterial sucrose-6-phosphate hydrolase.
