Effects of Bacillus subtilis JZXJ-7 From Shrimp Paste on Colonic Function and Microbial Community in Mice Infected By Vibrio parahaemolyticus

Abstract: The protective effects of Bacillus subtilis JZXJ-7 (JZXJ-7) on Vibrio parahaemolyticus (Vp) infected mice was investigated. Twenty-eight male C57BL/6J mice were randomly divided into four groups. The two pre-supplemented groups were given low and high doses of JZXJ-7 intragastrically for 14 d. The control group and Vp group were given equal volumes of normal saline every day. Standard virulent strain ATCC33847 was intragastrically given to 3 groups except the control group on day 15 (Vp infection murine model). The disease activity index (DAI), intestine (ileum, cecum and colon) Vp load, colonic pathological changes, colonic cytokine (interleukin-1β, interleukin-6 and tumor necrosis factor-α) levels, antioxidant enzyme (catalase superoxide dismutase, total antioxidant enzymes) activities and colonic microbial community were measured in mice. Compared with the Vp group, protective effect of the high dose (109 CFU) JZXJ-7 was > low dose, as evidenced by significantly decreased (P<0.05) DAI score, intestinal Vp load, inflammatory cell infiltration, collagen deposition, increased goblet cells number, inhibited cytokines-mediated inflammation, increased antioxidant enzyme activities and enhanced intestinal permeability in the colon. Additionally, JZXJ-7 markedly increased (P<0.05) microbial diversity, partially reversed the intestinal flora disturbances, enriched Akkermansia, Bifidobacterium,Clostridium and other beneficial bacteria, culminating in restoration of intestinal micro-ecological stability. Hence, JZXJ-7 effectively alleviates colonic pathological damage and stabilises the colonic function in Vp infected mice. These changes may mediated by microbial community changes.
