莲藕多酚萃取工艺的响应面法优化研究

Study on Optimizing Extraction Parameters of Lotus Root Polyphenol by Response Surface Methodology

摘要: 用乙酸乙酯作为萃取剂,从莲藕多酚提取液中萃取多酚、儿茶素,将响应面分析用于莲藕多酚萃取工艺条件的优化,建立了试验因素为pH(X1),相比(X2)和逆流萃取次数(X3)与萃取得率(Y1)和儿茶素百分含量(Y2)的动力模型。对萃取动力学进行了研究,莲藕多酚萃取最佳工艺条件pH为3.2,相比为1:1.5,逆流萃取次数为3。在最佳工艺条件下,多酚萃取物的含量是283.5mg/kg;而儿茶素萃取回归方程的各项回归均显著,说明还存在影响儿茶素萃取百分含量的其他因素,其最佳萃取条件有待于进一步研究。

Abstract: Polyphenol and catechin were extracted from lotus root by using acetic ester as solvent. The optimum conditions were obtained by the response surface methodology (RSM) tests. In this test based on three test factors, pH(X1), phase ratio (X2), countercurrent extraction times(X3), the dynamic models gave quantitative parameters for the extraction of polyphenol and catechin. The results showed that the optimal extraction conditions are as follows: pH3.2, phase ratio 1:1.5 and countercurrent extraction 3 times. Under these conditions the content of polyphenol is 283.5 mg/kg and regressions of the model of catechin extraction significant. The results indicated that there are other factors which can affect the content of the catechin, and further studies should be done later for the optimal conditions of catechin extraction.
