一株高产类胡萝卜素酵母Rhodotorrula.sp色素组分及摇瓶发酵研究

摘要: 对一株高产类胡萝卜素的红酵母Rhodotorula.sp的色素组分、生长及色素积累的动态变化和摇瓶最佳生理条件进行了研究。结果表明该株酵母色素含量高、色素主要成分为β-胡萝卜素;色素积累起始于对数期末期和稳定期初期;在本实验条件下,红酵母Rhodotorula.sp的最佳摇瓶发酵条件为蔗糖30g/L、硫酸铵10g/L、酵母膏3g/L、Mg-Cl20.3g/L、接种量30ml/L、恒pH6.0和培养基装量100ml/500ml;经72h培养其生物量、类胡萝卜素含量和产量分别达8.72g/L、1359.7μg/g和11.86mg/L。

Abstract: The carotenoid composition,courses of fermentation and optimum fermentation conditions were studied in carotenoid hyperproducing strain Rhodotoru sp,whose principal carotenoid pigment was β-carotenone.The pigments were accumulated at the end of exponential phase and the stationary phase.The cell biomass,carotenoid content and yield of strain Rhodotoru sp could reach 8.72g/L、1359.7μg/g and 11.86mg/L respectively under the selected optimum conditions. The optimum fermentation conditions for carotenoid production were as follows:sucrose 30g/L,(NH4)2SO410g/L,yeast extract 3g/L,MgCl20.3g/L,medium 100ml(in 500ml flask),steady pH6.0,inoculum size 30ml/L and culture time 72h at 28℃ and 150r/min.
