基于三重四级杆复合线性离子阱质谱法筛查和确证水产品中多种抗生素残留

Screening and confirmation of multiple antibiotic residues in aquatic products by triple quadrupole-linear ion trap mass spectrometry

摘要: 应用液相色谱-三重四级杆复合线性离子阱质谱(LC-Q-TRAP/MS)技术,建立了水产品中38种抗生素残留的快速筛查确证方法。水产品经酸化乙腈和乙二胺四乙酸二钠提取,浓缩后加正己烷去脂进行样品预处理。检测采用分段时间多反应监测-信息关联采集-增强离子扫描的方式(sMRM-IDA-EPI)及建立多种抗生素数据谱库,通过对化合物保留时间、离子对信息以及EPI谱图进行检索比对,完成了多种抗生素的筛查和确证,并采用内标法定量,实现了水产品中38种抗生素的同时定性和定量分析。结果表明,38种抗生素线性相关性良好(r > 0.99),方法的定量限在2.0 ~ 5.0 μg/kg,在10.0 μg/kg和50.0 μg/kg的添加水平下,平均回收率为63.5% ~ 113.7%,相对标准偏差为2.4% ~ 17.6%。本实验建立的水产品中抗生素药物残留的液相色谱-三重四级杆复合线性离子阱质谱分析方法,快速、准确、灵敏度高,定性能力强,可以成功应用于水产品中抗生素残留的筛查和确证分析。

Abstract: Rapid screening and confirmation method for 38 kinds of antibiotic residues in aquatic products was developed by high performance liquid chromatography-triple quadrupole-linear ion trap mass spectrometry(LC-Q-TRAP/MS).The sample of aquatic products was extracted with acidified acetonitrile and EDTA-2Na,then concentrated and purified by n-hexane.Finally,the samples were detected by LC-Q-TRAP/MS in multiple reaction monitoring with information-dependent acquisition of enhanced product ion(sMRM-IDA-EPI)mode followed with multiple antibiotic database.The 38 kinds of antibiotics were confirmed by retention times,ion pairs and the database searching using EPI library,and quantified by internal standard method.All the antibiotics showed good linearities with linear correlation coefficients all above 0.99.The limits of quantification(LOQs)for the antibiotics were 2.0 - 5.0 μg/kg.The average recovery for antibiotics ranged from 63.5% to 113.7% with the relative standard deviations(RSD)of 2.4% - 17.6% at spiked levels of 10.0 μg/kg and 50.0 μg/kg.The method is sensitive,accurate and can be applied to the screening of antibiotic residues in aquatic products.
