肠球菌素SAU-2 的纯化及其序列同源性分析

摘要: 对产自肠球菌SAU-2的肠球菌素SAU-2通过盐析、SP Sepharose Fast Flow阳离子交换层析纯化,并用Tricine-SDS-PAGE确定细菌素分子质量;同时分析肠球菌素SAU-2序列与已知细菌素序列的同源性。结果表明:肠球菌素SAU-2纯化倍数为10.79倍,回收率为17.27%;分子质量约为4350D;肠球菌素SAU-2是细菌素家族的新成员,其基因序列、氨基酸序列与多数已知的肠球菌素同源性较低。

Abstract: Enterococcin SAU-2 was partially purified by 90% ammonium sulfate precipitation followed by SP Sepharose Fast Flow chromatography. After the purification of enterococcin, a 10.79-fold enhancement in specific activity and a recovery rate of 17.27% were achieved. The estimated molecular weight of enterococcin SAU-2 was approximately 4350 D based on Tricine-SDS-PAGE. The structural gene and amino acid sequences of the enterococcin SAU-2 had low homology with those of most enterococcins. The enterococcin SAU-2 was identified as a new member of the bacteriocin family.
