固相萃取—C30—RPLC法测定南极磷虾油中的虾青素

Determination of Astaxanthin in Antarctic Krill Oil by C30 Reversed Phase High Performance Liquid Chromatography with Solid-Phase Extraction

摘要: 建立用固相萃取预分离、测定南极磷虾油样品中虾青素3种同分异构体的高效液相色谱法。样品经固相萃取小柱预分离净化,加NaOH-甲醇溶液皂化反应后,经YMC-Carotenoid C30色谱柱分离得到虾青素3种同分异构体,采用外标法定量。南极磷虾油中虾青素的最佳净化条件为:以LC-NH2为固相萃取小柱,正己烷-丙酮-甲醇(1:2:2,V/V)为洗脱溶剂,洗脱体积为8mL,洗脱流速控制在2~4mL/min。样品经LC-NH2固相萃取小柱净化后平均添加回收率为86.1%~94.3%,相对标准偏差为0.79%~1.91%。结果表明,方法重现性好,回收率高,适用于南极磷虾油中虾青素含量分析。

Abstract: An HPLC method for the determination of astaxanthin isomers in Antarctic krill oil was established following solid-phase extraction (SPE). Following clean up on LC-NH2 solid-phase extraction column, saponification of the extracted compounds was achieved with NaOH-methanol. Subsequent separation was performed on YMC-Carotenoid C30 column with n-hexane-acetone-methanol (1:2:2) as eluent at a flow rate between 2 and 4 mL/min. Injection volume was set at 8 mL. Quantification was performed by external standard method. Average recovery rates of astaxanthin isomers from spiked samples varied from 86.1% to 94.3% with RSD between 0.79% and 1.91%. Our results suggest that the developed method is of high reproducibility and recovery and suitable for the analysis of astaxanthin in Antarctic krill oil.
