天然类胡萝卜素抑制细胞恶性转化机理的初步研究

摘要: 实验对天然类胡萝卜素抑制细胞恶性转化的机理进行研究,结果表明: 斑蝥黄素、β-胡萝卜素和番茄红素均能显著升调细胞间隙连接通讯功能、抑制细胞生长,并使细胞周期时相左移、M期百分比下降、MI降低;细胞转化及类胡萝卜素处理过程中不伴随间隙连接蛋白基因Cx 43染色体DNA的丢失或突变。 提示: 类胡萝卜素抑制细胞增殖,升调细胞间隙连接通讯功能及其参入间隙连接蛋白基因Cx 43的表达与调控可能是其抑制细胞恶性转化的重要机制之一。

Abstract: The mechanism of natural carotenoids(including canthaxanthin, β-carotene, lycopene)on inhibiting DMBA-induced neoplastic tranformation has been investigated. The mouse embryo fibroblast C3H10T1/2 cell was used as cell model and the method of microinjecting fluoresence probe transfer was used for analyzing gap junctional intercellular communication(GJIC).The results showed that: carotenoids tested inhibited cell growth rate , proliferation and up-regulated GJIC siginificantly. There were no changes of chromosomal DNA of Cx 43 in different groups. It suggested that the effects of carotenoids on inhibiting proliferation and enhancing GJIC by regulating the protein gene Cx 43 might be one of the important mechanisms for inhibiting neoplastic transformation .
