灰树花多糖PGF-1体外对羟基自由基的抑制作用

摘要: 研究灰树花多糖PGF-1对脱氧核糖-铁体系产生的羟基自由基(•OH)的抑制作用;通过检测小鼠肝匀浆丙二醛(MDA)含量、肝线粒体MDA含量、肝线粒体肿胀度、肝线粒体膜流动性及大鼠红细胞氧化溶血,研究PGF-1体外对•OH引起的氧化损伤的保护作用。结果显示PGF-1具有明显抑制•OH的作用,可降低•OH所致MDA的产生、抑制小鼠肝线粒体的肿胀、对小鼠肝线粒体膜流动性具有保护作用,并对由•OH所致大鼠红细胞氧化溶血具有抑制作用。表明PGF-1体外对•OH有较强的抑制作用。

Abstract: The scavenging effect of PGF-1 (one kind of homogeneous polysaccharide from Grifola frondosa ) on hydroxylradicals (•OH) in deoxyribose-iron system was studied. To study the protective activities of PGF-1 against oxidation damagecaused by •OH in vitro, the content of malonaldehyed (MDA) in mice liver juice and mice liver mitochondria, the swellingextent of mice liver mitochondria, the mice liver mitochondria membrane fluidity and the hemolysis of rat red blood cell were assayed.Results showed that PGF-1 with obvious inhibitory effect on free radical of •OH. PGF-1 could decrease the formation of MDAinduced by •OH and inhibit the swelling extent of mice liver mitochondria. It also had protective activity on the mice livermitochondria membrane fluidity and inhibitory effect on the hemolysis of rat red blood cell induced by •OH. It suggested thatPGF-1 showed strong inhibitory effect on •OH in vitro.
