大豆异黄酮抑制大鼠乳腺癌细胞增殖的研究

摘要: 应用MTT法、3H-TdR掺入法及流式细胞术,研究了两种主要的大豆异黄酮棗大豆黄酮和染料木素对乳腺癌细胞增殖的抑制效应,并探讨了其作用机理。结果表明:两种大豆异黄酮均能够抑制乳腺癌细胞的生长,并呈明显的剂量和时间依赖效应;降低3H-TdR掺入量,抑制癌细胞DNA的合成;增加S期细胞的比例,阻滞细胞的周期活动;但未引起明显的细胞凋亡。

Abstract: To study the mechanism of growth inhibition of rat breast carcinoma cells by soybean isoflavones, cancer cell line was treated invitro with daidzein and genistein. Microculture tetrazolium(MTT)assay, 3H-TdR incorporation, and cell cycle flow cytometry were usedto study the inhibitory effects of daidzein and genistein on cell proliferation. Results showed that daidzein and genistein could result in adose-dependent and time-dependent growth inhibition and decrease 3H-TdR incorporation in DNA of rat breast carcinoma cell. Flowcytometry showed that cells were accumulating in the S phase, but cell apoptosis was not induced.
