黄皮新肉桂酰胺B的小鼠降脂活性及毒副作用

Study on the Toxicity and Hypolipidemic Effect of Lansiumamide B

摘要: 目的:首次探究鸡心黄皮果核的黄皮新肉桂酰胺B(Lansiumamide B, LB)作用于C57BL/6小鼠高脂肥胖模型中的降脂活性及在实验过程中的毒副作用,为其应用于更多活性领域研究提供安全性依据。方法:以C57BL/6小鼠为动物模型,经口灌胃LB以探究对小鼠的NOAEL(No Observed Adverse Effect Level),作为降脂实验的浓度设置依据;建立高脂小鼠模型后,对小鼠给药灌胃4周,通过体重、血脂生化指标、胰岛素抵抗、氧化应激等指标评价LB的降脂活性。结果:经口急性毒性实验结果得出LB对C57BL/6小鼠的NOAEL为2 g/kg·bw (body weight);高脂模型实验结果显示LB能够显著降低小鼠体重,HLB(80 mg/kg药物组)与HFD(高脂模型组)相比:体重下降15.51%,Lee's指数下降3.25%,血清中HDL-C(High Density Lipid-Cholesterol)上升38.04%,SOD(Superoxide Dismutase)、GSH-Px(glutathione peroxidase)和CAT(Catalase)酶活提高了41.86%、41.95%和105.50%。结论:LB的NOAEL为2 g/kg·bw,在NOAEL内的浓度对小鼠具有显著降脂活性且无明显且无可见毒副作用,为开发其成为天然降脂药物提供理论依据。

Abstract: Objective: To explore the hypolipidemic activity of lansiumamide B (LB) based on the high-fat-diet model in C57BL/6 mice for the first time. Methods: The C57BL/6 mice were intragastric administrated with LB to explore the NOAEL (no observed adverse effect level) of LB. The concentration of the lipid-lowering experiments were set on the basis of the NOAEL of LB. After establishing the high-fat model, mice were intragastric administered for 4 weeks, then evaluated via body weight, biochemical indexes, insulin resistance, major organs injury and skeletal development. Results: Through the acute oral toxicity experiment results, the NOAEL of LB on C57BL/6 mice was 2 g/kg·bw (body weight). The results of the hypolipidemic experiment suggested that LB significantly reduced the body weight of mice. Among of these results, compared to HFD (high-fat-diet model group), the body weight of HLB (80 mg/kg drug group) reduced 15.51%, the Lee's index fell 3.25 %, HDL-C (High Density Lipid-Cholesterol) increased by 38.04%, the SOD (Superoxide Dismutase), GSH-Px (glutathione peroxidase) and CAT (Catalase) enzymes increased by 41.86%, 41.95% and 105.50% respectively. Conclusion: The NOAEL of LB is 2 g/kg·bw and the concentration within the NOAEL has observably hypolipidemic activity in mice.
