Partial Characterization and Antioxidant Activity of Water-soluble Polysaccharide Isolated from Bulbus Fritillariae ussuriensis

Abstract: A water-soluble polysaccharide was isolated from Bulbus Fritillariae ussuriensis (dried bulbs of Fritillaria ussuriensis Maxim) sequentially through water extraction, ethanol precipitation, ion exchange chromatography and gel permeation chromatography. The molecular mass of FUP-1 was determined by gel filtration chromatography, and its structure was characterized by HPLC and FTIR spectroscopy. The results showed that FUP-1 was a heteropolysaccharide, of which the monosaccharide composition was composed of xylose, glucose and galactose with a molar ratio of 5:1:1. The average molecular weight of FUP-1 was determined to be 4.1 × 104 D. Bioactivity tests in vitro showed that FUP-1 displayed strong antioxidant effect and scavenging capacity against hydroxyl, DPPH, and superoxide anion free radicals. Therefore, FUP-1 may be a potent antioxidant agent.
