Study on Preparation of High-Intensity Sweetener-Trideoxygalactosucrose by Monoesterification

Abstract: Preparation of high intensity sweetener 4,1’,6’-trichloro-4,1’,6’-trideoxygalactosucrose by using monoesterification has been studied.The 6-position hydroxyl group in sucrose was protected by the reaction of sucrose with triethyl orthoacetate.Sucralose was prepared by chlorinated sucrose 6-acetate with thionyl chloride/pyridine.The optimum chlorination condition was:sucrose 6-acetate syrup(20g),Thionyl chloride(20~22ml),1,2-dichloroethane as reaction mediums and refluxed time was 10~11 hours.The chlorinated product was hydrolyzed with methanol-sodium methoxide to deacetylation.With the mono-esterification,the preparation route should be simple and the reaction condition was more gentle than with tritylation.
