Purification and Physico-chemical Characteristics Analysis of D.fordii Prain et Burkill Fraction DFPN-Ⅰ

Abstract: The crude polysaccharide was hot water extracted from D. fordii Prain et Burkill, and precipitated by ethanol. Apolysaccharide named DFPN-Ⅰ were prepared after degreasing ,deproteinixation and decoloring, and further purified on two timesSephadex G-75 column chromatography. Homogentity was examined with paper chromatography and determination of specificrotatory power and finally identified by its molecular weight. DFPN-Ⅰwas a non-starch polysaccharide without any protein,nuclear acid, polyphenol and uronic acid. Its average molecular weight was 48200. The unit of the monosaccharide was identifiedby paper chromatography and gas chromatography as mannose, glucose, gala-ctose with a mole ratio of 1.26:1:2.87 respectivelyafter complete hydrolysis by acid.
