Study on Solid Phase Extraction and Spectrophotometric Determination of Lead in Biological Sample with 2-Carboxyphenylazo-Rodanine

Abstract: The color reaction of lead with 2-carboxyphenylazo-rodanine (CPAR) was studied. In the presence of pH4.0 buffer solution and emulsifier-OP medium, CPAR could react with lead(II) to form a stable 2:1 complex. This complex extracted by Waters Sep-Pak-C18 cartridge and eluted by DMF, its spectrophotometry determination results showed that the molar absorptivity is 6.75×104 L/mol·cm at 520 nm. Beer’s law was obeyed in range of 0.05~4 μg/ml. This method can be applied for the determination of lead in biological sample with satisfactory results.
