Determination of Carotenoids in Tuber Flesh by HPLC in Potato(Solanum tuberosum L.)

Abstract: A high performance liquid chromatographic method was used to determine carotenoids in potato tuber flesh. Carotenoids were extracted from tuber flesh with acetone and petroleum ether(1:1, contain 0.1 % BHT). Using ZOBAX C18 column (4.6×250 mm, 5μm), acetonitrile:methanol:methylene chloride (75:20:5, contain 0.1 % BHT) as mobile phase, carotenoids were eluted and monitored at 450 nm.The method showed good linearity (the correlation coefficient were 0.9997 and 0.9987) and repeatability.The recoveries of lutein and zeaxanthin were 92.87 % and 95.68 %, and oxidation of carotenoids was effectively prevented.It proved to be a very useful method for testing large quantities of samples.
