Study on Optimization of Extraction of Red Pigment from Jujube Peel with Ultrasonic-assisted Cellulase-Alkaline Solution by Response Surface Analysis

Abstract: In this study, the optimal conditions of cellulase reaction for destroying cell wall of jujube peel were determined by response surface analysis (RSA), and extraction conditions of pigment with alkaline solution were optimized by orthogonal test. The optimal parameters of cellulase reaction are quality of cellulase 1.26%, enzyme reaction temperature 51.90 ℃, pH value 5.19, and enzyme reaction time 60 min. And the optimal parameters of extraction with alkaline solution are alkaline solution concentration 1.0%, extraction temperature 80 ℃, extraction time 35 min. Under these conditions, the extraction rate of red pigment is up to 1.326μg/g.
