Structural modification and solubility enhancement of Glutenin by pH-migration

Abstract: The unique amino acid composition of Glu leads to its poor solubility, which greatly limits its application in the food industry. The pH shift treatment can induce structural changes of protein and significantly improve the solubility. Therefore, Glu was modified by pH shifting to improve its properties in this paper. The effects of pH migration treatment on the properties of glutenin were investigated by dynamic light scattering, TEM, fluorescence, FT-IR dichroism. The results showed that pH migration could significantly reduce the particle size of Glu aggregates and increase the surface charge of Glu granules. Increase the hydrophobicity and sulfhydryl group content, decrease the disulfide bond content, the effect of alkali migration was more significant than that of acid migration. In addition, base migration treatment significantly improved the solubility of Glu and play an active role in broadening the application range of Glu in food industry.
