响应面法优化山麻楂粗多糖提取工艺

Optimization of Extraction Process for Polysaccharides from Stems and Leaves of Gypsophila oldhamiana Miq. by Response Surface Methodology

摘要: 用响应面法优化野生山麻楂粗多糖的提取条件。在单因素试验基础上,选择提取温度、提取时间、液料比三因素三水平进行Box-Behnken试验,确定提取工艺的优化组合条件。结果表明:提取工艺最佳条件为提取温度97℃、提取时间3.5h、液料比30.5:1(mL/g)、提取2次,该条件下多糖得率预测值为2.226%,验证值为2.219%。

Abstract: Response surface methodology was employed to optimize conditions for the extraction of polysaccharides from the stems and leaves of wild Gypsophila oldhamiana  Miq. On the basis of single-factor experiments, temperature, time and  solvent-to-solid ratio were selected out of four extraction conditions for a three-level Box-Behnken experimental design. Results showed that optimal extraction conditions were obtained using two extraction cycles at 97 ℃ with distilled water at a solvent-to-solid ratio of 30.5:1 (mL/g) for 3.5 h. Under these conditions, the predicted and experimental extraction yields of polysaccharides were 2.226% and 2.219%, respectively.
