响应面法优化紫山药花青苷提取工艺及抗氧化性的研究

The response surface method to optimize the purple yam anthocyanins and the oxidation resistance research

摘要: 以紫山药为实验材料,以酸性乙醇为提取溶剂通过Box-benhnken响应面法及Design-Expert 8.0.6分析软件建立二次多项式数学模型优化紫山药花青苷的提取工艺及参数。同时,对紫山药花青苷清除羟基自由基和超氧阴离子的能力进行了抗氧化研究。研究表明,五种单因素对花青苷得率影响大小的顺序为X5(盐酸质量分数)> X2(时间)> X4(乙醇体积分数)> X3(料液比)> X1(温度)。得到紫山药花青苷最佳提取工艺参数为温度:80℃、时间:3.5h、料液比:1:25、乙醇体积分数:70%、盐酸质量分数:18%。在此条件下花青苷平均得率达到4.966mg/g,相对标准偏差为0.29%,与数学模型理论得率的相对误差小于1.0%。抗氧化性实验表明,紫山药花青苷对?OH及O2-?具有较好的清除能力,其抗氧化能力强于Vc。

Abstract: In this paper,With purple yam as experimental material, extracted by acidic ethanol as the solvent through the Box - benhnken response surface method and the Design - Expert 8.0.6 analysis software to establish the mathematical model of quadratic polynomial optimization of the extraction process of purple yam anthocyanins and parameters. At the same time, the purple yam anthocyanins’ ability to remove hydroxyl radicals and superoxide anion was studied in the resistance of oxidation. Research shows that five kinds of single factor to anthocyanins yield affecting the size of the order for X5 (mass fraction of hydrochloric acid) > X2 (time) > X4 (volume fraction) ethanol > X3 (material liquid ratio) > X1 (temperature). Get purple yam anthocyanins’ optimal extraction process parameters are temperature: 80 ℃, time: 3.5 h, solid-liquid ratio: 1:25, ethanol volume fraction: 70%, hydrochloric acid mass fraction: 18%. Under the condition of the anthocyanins average yield of 4.966 mg/g, relative standard deviation was 0.29%, and the mathematical model between the theoretical yield, the relative error less than 1.0%. Antioxidant activity of the experimental results show that the purple yam anthocyanins have a good ability to remove? OH and O2 - ? and its antioxidant ability is stronger than Vc.
