榛仁分离蛋白降脂活性肽分离纯化及结构鉴定

摘要: 摘 要:采用超滤、Sephadex G-25、Sephadex G-15、反相高效液相色谱(RP-HPLC)及质谱对榛仁分离蛋白降脂活性肽进行分离纯化及结构鉴定,并通过测定3T3-L1前脂肪细胞诱导分化过程中脂质积累、总胆固醇及甘油三酯水平,筛选出具有较高降脂活性的多肽氨基酸肽段。结果表明,经Sephadex G-15分离得到的C3组分的胰脂肪酶抑制、胆固醇胶束吸附及细胞降脂活性均显著高于其它组分。进一步经质谱解析筛选出的肽段Phe-Leu-Leu-Pro-His(FLLPH)与模型组相比,可抑制26.31%的总脂形成,降低32.67%胆固醇和23.87%甘油三酯水平。FLLPH具有较好的降脂活性,本研究可为榛仁降脂活性肽的开发提供理论参考。

Abstract: Abstract:Hypolipidemic activity peptides of Hazelnut isolate protein were separated and purified by ultrafiltration, Sephadex G-25, Sephadex G-15 and reversed-phase high-performance liquid chromatography (RP-HPLC). The amino acid structural of peptides with the best hypolipidemic activity were identified by Q Exactive hybrid quadrupole-Orbitrap mass spectrometry, and the levels of lipid accumulation, total cholesterol as well as triglyceride in the 3T3-L1 preadipocyte were also determined. The results showed that the C3 fraction isolated from Sephadex G-15 had significantly higher pancreatic lipase inhibition, cholesterol micelle adsorption, and cell hypolipidemic activity than other fractions. The peptide Phe-Leu-Leu-Pro-His (FLLPH) selected by mass spectrometry analysis could inhibite 26.31% of total lipid formation, and reduce 32.67% of cholesterol and 23.87% of triglyceride levels compared to the model control. The present study evaluated the hypolipidemic potential of FLLPH, and provided a theoretical reference for the development of hazelnut hypolipidemic active peptide.
