一种RNA提取试剂盒——TRIZOL的使用方法初探

摘要: 以酿酒酵母为原料,利用TRIZOL试剂快速提取法,得到的总RNA不完整,只有5srRNA一条条带,经过对该试剂盒的改进后提取的总RNA呈现28srRNA,18srRNA和5srRNA三条清晰的条带,很少有降解,其A260/A280值可达1.946,A260/A230值为2.070,具有很高的纯度。

Abstract: To isolate total RNA fromS accharomyces cerevisiae by the method of TRIZOL reagent kit, the RNA purified was rather not complete, since it showed band 5s rRNA only. After changing several steps of the method, the results showed that there appeared three clear bands—28s rRNA, 18s rRNA and 5s rRNA respectively with little RNA decomposed. The ratio of A260/A280 was 1.961, and A260/A230 2.070, so the purity was high.
