黄粉虫中提取纯化超氧化歧化酶的工艺研究

摘要: 本研究以黄粉虫粉为原料,经盐析沉淀,DEAE-Sepharose FF阴离子交换柱层析和Sephadex G200凝胶过滤层析等纯化步骤,获得了其中的一种超氧化物歧化酶(SOD)。酶的纯化倍数为78.3倍。SDS-PAGE电泳显示为均一的蛋白谱带。经鉴定该酶为Cu.Zn-SOD。本研究为从黄粉虫中提取SOD提供了一种较好的工艺。

Abstract: In this paper,a kind of superoxide dismutase(SOD)was obtained from yellow worm powder.Two-step(NH4)2SO4 precipitation were used as the main extraction method and DEAE-Sepharose FF ion-exchange and Sephadex G200 volume exclude chromatography were used as purification methods.The purification factor was 78.3.The SDS-PAGE showed that the SOD was a homogenous protein.This enzyme was proved as Cu.Zn-SOD.This study provides a suitable technology for obtaining SOD from the yellow mealworm.
