油松花粉多糖提取及其清除羟自由基活性研究

Study on Extraction and Hydroxyl Radicals Scavenging Activity of Polysaccharides from Chinese Pine Pollen

摘要: 通过单因素试验和正交设计试验,研究油松花粉多糖水提取条件。结果表明:提取温度和提取时间是影响多糖提取得率的主要因素,最佳提取条件为:料液比1:25,90℃水浴条件下浸提,每次浸提3h。在此条件下油松花粉多糖提取得率达1.4%;另外采用水杨酸法检测油松花粉多糖对羟基自由基清除作用。结果表明油松花粉多糖对羟基自由基有较明显的清除能力,清除率达50%,所需多糖浓度为0.75mg/ml。

Abstract: The water extraction conditions of polysaccharides from Chinese pine pollen were optimized by single factor test and orthogonal design. Results showed that extraction temperature and extraction time are the major factors affecting the polysaccharides extraction rate. The optimal extractions conditions are as follows:extraction temperature 90 ℃,extraction time 3 h and ratio of material to water 1:25. In addition,the antioxidant activity of the Chinese pine pollen polysaccharides was evaluated by(·OH) free scavenging assay. Results indicated that the polysaccharides show strong hydroxyl radicals scavenging activity and the 50% effective concentration(EC50) is 0.75 mg/ml.
