翡翠贻贝粗多糖降血脂作用的研究

Hypolipidemic Effect of Crude Polysaccharides from Perna viridis

摘要: 目的:研究翡翠贻贝粗多糖(PCP)对高脂血症小鼠的降血脂作用及其机制。方法:取雄性昆明小鼠72只,每组12只。随机分正常对照组(灌饲生理盐水)、模型对照组(灌饲生理盐水)、阳性对照组(灌饲辛伐他汀5mg/kg) 、翡翠贻贝粗多糖低剂量组(50mg/(kg·d))、中剂量组(100mg/(kg·d))、高剂量组(200mg/(kg·d))。正常对照组小鼠从实验开始至结束用普通饲料喂养,其他5组小鼠均喂高脂饲料。30d后测定小鼠血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)和一氧化氮(NO)含量,肝脏TC、TG含量,血清卵磷脂胆固醇酰基转移酶(LCAT)以及血清和肝脏脂蛋白脂酶(LPL)、肝脂酶(HL)活性。结果:翡翠贻贝粗多糖能明显降低模型小鼠血清TC、TG、LDL-C含量和动脉粥样硬化指数(AI)(P<0.01),显著降低肝脏TC含量(P<0.01)、TG含量(P<0.01),提高血清HDL-C含量和NO含量(P<0.01),提高血清LCAT、血清和肝脏HPL、HL活性(P<0.01)。结论:翡翠贻贝粗多糖对小鼠有显著的降血脂效应,该作用可能与翡翠贻贝粗多糖能明显增强LCAT、LPL、HL酶活性,提高HDL-C、NO水平有关。

Abstract: Objective: To investigate the hypolipidemic effect of crude polysaccharides from Perna viridis (PCP) in hyperlipidemic mice and its mechanisms. Methods: Totally 72 Kunming male mice were arbitrarily divided into normal control group, hyperlipidemia model group, positive control group, PCP low-dose group, PCP medium-dose group, and PCP high-dose group. The normal control group was fed normal diet and other five groups were fed high-dat diet. The normal control group and model control group were administered with the same volume of normal saline and the positive control group with simvastatin tablet at the dosages of 5 mg/kg. The low, medium and high dose groups were administered with crude polysaccharides from Perna viridis at the dosages of 50, 100 mg/kg and 200 mg/kg, respectively. All the administrations were carried out once a day for 30 days.  The contents of serum total cholesterol (TC), triglyceride (TG), low density lipoprotein-cholesterol (LDL-C), high density lipoprotein-cholesterol (HDL-C) and NO, the contents of liver TC and TG, and the activities of serum lecithin cholesterol acyltransferase (LCAT) as well as lipoprotein lipase (LPL) and hepatic lipase (HL) in the serum and liver were determined after the last administration. Results: Crude polysaccharides from Perna viridis could decrease the contents of serum TC, TG and LDL-C and AI, increase the contents of serum HDL-C and NO, reduce the contents of liver TC and TG, and enhance the activities of serum LCAT and the activities of HPL and HL in the serum and liver. Conclusion: Crude polysaccharide from Perna viridis can significantly decrease blood lipid and the mechanism may be related to the improved activities of LCAT and LPL and increased levels of HDL-C and NO.
